I joined the Newcastle 2018 iGEM team as part of my MSc Thesis. As part of GeneMachine, a post-graduate subgroup of the Alternative Roots Newcastle iGEM Team, we investigated the issues of irreproducibility and standardisation in Synthetic Biology. More specifically we focused on the 2018 Interlab and what could be done to improve it. My aim was to apply a Bio-design Automation methodology that used liquid handling robots and Design of Experiments (DoE) to rapidly optimise and generate competent Escherichia Coli DH5-alpha. These could than be used for automated transformation protocols that would allow for the high-throughput, lab-scale transformation of ultra-competent E. coli. The liquid handling robot used was the recently released Opentrons OT-2, a robot we won in an international competition, the only team in the UK to do so. This project required me to be highly interdisciplinary, learning how to code in python and working alongside architects and engineers to construct hardware to compliment the software. Alongside this I also helped the Alternative Roots team to use laboratory robotics to aid in the automation of their experiments.

I also took an optional module in Ideas for Business Enterprise in STEM.I undertook an 8 week molecular biology practical skills workshop that cumulated in the use of ligation independent cloning to generate a recombinant bacteria which produced mCherry fluorescent protein. From this immobilised affinity chromatography (IMAC) and western blots were used for analysis protein presence. I also undertook a research based bioinformatics module that used DNA and protein databases to discover a chitinase enzyme that may be useful in an industrial setting. Phylogenies were constructed, as well as explicit protein models using Chimera.
