Investigated how HSV-1 regulates the host type-I interferon response by high-throughput analysis of the viral genome• Designed a novel transposon construct that allows for high-throughput sequencing of pooled viral libraries• Constructed a viral library of HSV-1 by transposon mutagenesis • Screened the viral library in WT and IFNAR1-/- human cell lines, thereby identifying novel viral regulators of IFN-I• Uncovered the molecular mechanism by which the viral protein UL42 inhibits interferon induction2.
