Project 3: Studies on the mechanism of Bortezomib (Velcade®) resistance in preclinical models of multiple myeloma.Bortezomib (Velcade TM) is considered to be one of the first and better proteasome inhibitor for treatment of Multiple Myeloma (MM) approved by FDA. This drug is very effective in treatment of patients with multiple myeloma who have failed in prior therapy. In new diagnosed MM patients a combination of Bortezomib, thalidomide and dexamethasone showed effective response. But in certain Human Myeloma Cell Lines (HMCL) showed resistance to bortezomib (b). Attempts are made to understand the signal transduction and it’s up-regulatory and down- regulatory elements at the gene level. At Dr. Wang’s Laboratory; performed cloning, transformation and purification of recombinant proteins (prokaryotic system) used as substrates for in-vitro ubiquitination assay. With the aid of in-vitro ubiquitination assay attempts were made to purify the novel E3 ligases from the nuclear and cytosolic extracts from various distinct cell lines using conventional protein purification techniques (FPLC).
