As an undergraduate Research Assistant in Buttitta Lab, MCDB department I examined chemotherapy drugs, Docetaxel and GM-CSF effects on disseminated tumor cells (DTCs) and the decision to enter quiescence or the cell cycle. Cells were innately marked with different cell cycle reporters and live imaged. These images were then spliced together to create “cell movies” where we could observe single cells and their entry or exit into the cell cycle. While studying this decision to enter the cell cycle, I initially utilized a FUCCI-based reporter system however cells lost markers before our imaging time was concluded. To correct this, we utilized the endogenous fluorescent Tagging Toolkit (eFlut) technique. The eFlut assisted in creating labeled cell lines for our further studies. Eventually, our research expanded to pathway studies. The Hippo signaling pathway is key in inhibiting uncontrolled cell growth. I was able to create Tet-inducible cell lines for Hippo tumor-suppressing signal pathway components YAP and TEAD2 for examining their effects on the proliferation-quiescence decision in culture. I utilized the following methods: Cell culture, confocal microscopy, Drosophila technique (genetics, dissection via light microscopy, etc.), PCR/, gel electrophoresis, CRISPR-Cas9, transfection, cell line generation (eFlut), DNA extraction/purification
