About Nicolas Sévère
I am a dedicated biomedical scientist and expert in stem cell biology and oncology, with unique insight in bone biology. Using cutting-edge biology techniques, I deciphered the complexity of the bone marrow microenvironment at single-cell resolution and together with in-vivo models I identified subsets of stromal cells regulating hematopoietic cells regeneration post transplantation, as well as their contribution to bone metastases development. I am now looking for a new challenge and for using my skills to develop new therapies for patients.Skills:• Unique expertise in mesenchymal stromal cells, bone tropic cancer cells, hematopoietic stem cells• Mass cytometry (CYTOF) and single-cell RNA sequencing sample preparation from in-vivo models and human tissues• Ability to develop projects in a collaborative manner demonstrated by co-authorship of 12 publications with scientists of my workplace and other labs, highlighting my team spirit• Key protein classes studied: Ecto-5’nucleotidase (CD73), E3 ubiquitin ligase (cbl-c), Tyrosine Kinase Receptors• Supervising and mentoring undergraduates; organizing lab summer student programs

I worked in the Scadden lab in the Center for Regenerative Medicine where I complemented my understanding of bone biology with the hematopoietic niche dimension. My main interest was to define the heterogeneity of bone marrow stromal cells within the hematopoietic niche at single-cell protein level using a novel high parameter technique, Mass Cytometry. Notably, I defined a new subset of CD73+ stromal cells participating to acute blood regeneration following bone marrow transplantation. Also, I had the chance to initiate several collaborations with labs working in different biomedical fields and successfully applied my bone marrow niche expertise to support their research.

I worked under the supervision of Dr Pierre Marie in the Osteoblast Biology and Pathology Team, in the Bones and Joints lab, INSERM U606/University Paris Diderot. My interest was the role of E3 Ubiquitin ligase Cbl in bone pathologies and my 2 major projects were i) to study of function of Cbl during Mesenchymal Stromal Cells differentiation into osteoblasts and ii) to characterize the function of Cbl in bone cancer formation (osteosarcoma).
