DMPK project lead on an internal early discovery program where I provided guidance on in vitro and in vivo study design including in vitro ADME assays (including hepatocyte/microsome/plasma stability and protein binding) and in vivo PK.Designed and conducted CYP450 induction assay using cryopreserved hepatocytes and suitable LC-MS/MS analytical methods for FDA recommended probe substrates for CYP1A, CYP2B, and CYP3A, and used these data to assess compounds in early discovery to help understand potential DDI liabilities. Added secondary cell viability endpoint (Cell Titer Glo) to the assay in order to increase utility of each experiment.Assessed direct and metabolism-dependent CYP450 inhibition of proprietary compounds. Also performed assay development for the assessment of CYP2C8 inhibition.Performed in vitro experiments in support of drug discovery including compound metabolic stability (suspension hepatocytes, microsomes), protein binding, and plasma stability.Brought BSEP vesicle-based transporter assay in-house and developed LC-MS/MS analytical methods.Pharmacokinetic study design input, sample analysis, and data interpretation in various matrices (including plasma, lung, liver, tumor tissue) in support of multiple early discovery programs. Data analysis performed using Analyst and Phoenix WinNonLin.Performed compound optimization, method development, mobile phase preparation, sample analysis, and system maintenance/troubleshooting on LC-MS/MS systems.
