About Peter Clements
My PhD at the University of Adelaide was on the active site of the enzyme pyruvate carboxylase. Then followed two postdoctoral fellowships in the USA at the University of Wyoming and then at Johns Hopkins University school of medicine in Baltimore Maryland. Returning to Adelaide I took up a position as a principal medical scientist at the Women’s and Children’s hospital and was there for 30 years until retirement in February 2011. I worked in a research group of up to 35 people on a group of lysosomal diseases known as the mucopolysaccharidoses. I began by purifying the normal human enzymes from liver (obtained from the forensic department). We then sequenced enough of the enzymes to get a DNA probe from which we isolated the genes. We then expressed the genes in CHO expression lines and obtained enough purified enzyme to begin trials in a cat model of one of the diseases. Having determined dosages we then formed commercial partnerships and produced the enzymes commercially for worldwide distribution to patients. At the same time we were able to purify enough of the enzyme we were treating the cats with to obtain a crystal structure. The first structure obtained of a sulphatase class of enzymes. We then set up phase II and phase III trials in patients who tested positive for deficiency of the enzyme. The enzymes are now used worldwide, which took 25 years after the initial research began. The sales bring back millions in royalties to the state health dept. Once treatments became available it became necessary to develop early screening systems and I then became involved in mass spectrometric methods to screen for these diseases. We ran a diagnostic service for these diseases which serviced all of Australia, New Zealand, some South East Asian centres, Saudi Arabia & Qatar . My FHGSA is a professional qualification, fellow of the human genetics Society of Australasia in biochemical genetics, was awarded by the Human Genetics Society of Australasia in 2000.

We are looking to extend the research into analysis of fatty acids in blood spots taken from infants and this will require the extra sensitivity of mass spectrometry. Thus far there is a strong interest.
