My PhD project focused on exploring polyclonal antibody responses induced by Klebsiella pneumoniae outer-membrane vesicles (OMVs). In brief, the project involved the production of OMVs before extensively characterising them, examining size, concentration, morphology and protein content. OMVs were then used to immunise mice, producing sets of polyclonal serum. Each set of serum was then characterised. Determining key antigenic targets, dominant antibody isotypes, and cross-binding potential against OMVs from different isolates. I also employed high content imaging as a method to determine antibody cross binding potential against a collection of 175 different characterised K. pneumoniae isolates before then investigating potential functional responses including complement deposition and opsonophagocytic.As of February 2024 I had submitted my PhD thesis with a viva date pending. The research involved working with patient samples measuring key biomarkers using a mixture of ELISA, Luminex technology and RT-PCR. These data were then combined with existing patient metadata to try and understand associations between obesity and serve dengue fever in a matched cohort study. This role involved handling large and complex datasets, teaching myself how to work with a new coding language (R), and working across cultures and language barriers.
