I studied the function of the putative RNA Helicase HELZ using molecular biological, biochemical and immunological tools. The following are my main scientific achievements:1. The putative RNA helicase HELZ promotes protein translation and cancer cell proliferation.• I found that depletion of HELZ from MCF7 (human breast adenocarcinoma) cells or HeLa (cervical cancer) cells results in a significant decrease of cell proliferation. • HELZ plays a role in translation initiation and the phosphorylation of the ribosomal protein S6, which is a component of the mTOR signaling pathway. This pathway is known to be dysregulated in many human cancers. 2. Conserved LxxLAP motifs within a protein are not a prerequisite for oxygen-dependent regulation.• Even though the site of oxygen-dependent regulation in the HIF protein is found within a LxxLAP protein sequence motif, in other proteins it is not. Therefore, a protein sequence screen for this motif does not help to find novel proteins that play a function within the hypoxic microenvironment of a tumor.
