Supported by the results of X-ray analysis of inhibitor: kinase co-crystals and extensive structure-affinity studies enabled to constructed inhibitors whose affinity reached picomolar range. I was introduced to new methods for the synthesis of complicated heteroaromatic systems, bicycles, and tricycles. These aromatic fragments were conjugated with peptide analogs on solid phase and labeled with fluorescent dyes. The obtained molecular conjugates possessed unique photoluminescent properties and they were used as optical probes for mapping of localization and monitoring activity of protein kinase in cells, but also HTS of inhibitors of protein kinases in biochemical binding/displacement assays. Further, he worked with photoluminescence (fluorescence and phosphorescence) based biochemical binding/displacement assays, using detection systems based on luminescence intensity, fluorescence polarization/anisotropy, time-gated luminescence intensity, and luminescence lifetime measurement. The aim of these measurements was the development of technologies for disease biomarker analysis. Show less
