About Ronald Etheridge
The vast majority of my academic career has been devoted to the study of human parasites with the goal of elucidating the molecular machinery behind host-parasite interactions. This work has allowed me to develop a comprehensive set of technical skills and problem solving strategies required for cutting edge basic research applications.Apart from my technical abilities listed below, I enjoy work in collaborative groups and have been involved in many successful collaborative projects during my graduate and post-doctoral training with several co-first author manuscripts demonstrating this capacity.Technical Skills:• Generation of vectors for use in prokaryotic and eukarytic hosts for use in gene expression, tagging, and knockout strategies including CRISPR/Cas9 methods. Trained in restriction digest vector construction, Gibson Assembly cloning, TOPO and Gateway cloning methods.• Use of gravity flow and AKTAexplorer platform FPLC methods of protein purification.• Running of SDS/Native-PAGE/Agarose gel based separation of proteins followed by Western or Far-Western blotting for downstream analysis.• Generation, isolation and manipulation of RNA using a variety of methods. Utilized techniques such as Northern blotting, radiolabeled RNA sequencing gels, cDNA production and qRT-PCR.• Extraction/isolation of DNA from a variety of sources and analysis with including Southern blotting, EMSA’s and ChIP.• Performed experiments involving mouse infection studies and analyzed mice using blood smear or luciferase IVIS imaging. Produced and tested mouse polyclonal antibodies to proteins specific to my research projects.• Maintained mouse colonies and the breeding and weaning of pups.• Familiar with BioPlex, Flow Cytometry and Single Cell Sorting • Proficient at general techniques of fixation, permeabilization and staining with both dyes and antibodies.• Experienced in the sample preparation and machine operation of the JEOL 1200 EX II Transmission Electron Microscope.

Projects included the systematic knockout of each member of the highly expanded secreted kinase family (~16 members) and the phenotypic characterization of the resulting knockout parasite.• Novel tandem affinity purification strategies were implemented in T. gondii in order to provide the first identification of components of rhoptry kinase complexes using mass spectrometry and verified through reciprocal tagging. • Additional collaborative projects focused on identifying and characterizing parasite effectors responsible for suppression of the STAT1 pathway in infected cells as well as elucidating the mechanism behind effectors involved in subverting host cell machinery involved in destruction of the parasite vacuole. • Projects included the characterization of novel non-canonical poly(A) polymerases present in the human pathogen Trypanosoma brucei and Leishmania major. This work provided the first mass spectrometry based analysis of a mitochondrial poly(A) polymerase. Additionally my work focused on the characterization of distinct protein complexes involved in RNA processing events within the mitochondria of these organisms.
