The Brussels Liver Research to Valorisation Alliance (B-Liver) aims to integrate the complementary expertise of VUB’s liver research units to capitalize on common valorisation pathways that transect existing academic liver and safety research lines. B-Liver’s mission is grounded in proven expertise in liver cell biology, pathology and toxicology, which enables the consortium to also extend into higher-risk emerging innovations::: HepaSCREEN :: Builds on a diverse pool of liver based in vitro models, encompassing various cell types, from multiple sources, in different configurations. The common goal between models is to develop better models and tools for toxicity and disease prediction and to develop new therapeutic strategies for liver pathology.:: EvoMED :: Here B-Liver is developing a pipeline of innovative technologies that can fill therapteutic needs in the field of inherited metabolic disorders. :: Public Health and 3R AMs :: The toxicology and 3R aspects of the consortium’s activities also finds impact in a societal context. To this end, the need to ensure safety for both patients and consumers, in addition to regulatory drivers, underlie the public health aspects of B-Liver.

Retrospective analyses show that safety and efficacy constitute major stumbling blocks during drug development. In particular, drug-induced liver injury (DILI) is the main safety-related reason of post-marketing drug withdrawal and the major cause of acute liver failure in the Western world.The inability of currently available preclinical animal-based safety studies to predict the hepatotoxic potential of new therapeutic agents is one of the underlying causes. Additionally, animal tests are time- and resource-intensive, and remain subject of ethical controversy. The current European regulatory environment also stimulates the shift from in vivo to in vitro testing, in particular because the use of animals for research purposes becomes increasingly prohibited.Consequently, not only the pharmaceutical, but also the chemical and cosmetic industries urgently need in vitro models for toxicological investigations. These should be:(i) human-relevant,(ii) adequately reflect specific tissue function, and,(iii) enable process automation and high-throughput.The In Vitro Toxicology team strives to find an answer to these needs by developing metabolically-competent in vitro liver models based on: - Primary hepatocytes- Stem cells- Connexin-based cellular signalling in liver disease- Identification of genotoxic compounds using a human hepatoma cell-based classifier
