I am an enthusiastic doctoral candidate in the Nanoscience & Microsystems Engineering program at the University of New Mexico (UNM). My Ph.D. research under the supervision of Dr. Keith Lidke (Professor, Department of Physics & Astronomy) is focused on the development of single molecule localization microscopy (SMLM) methods and their data analysis. The title of my dissertation is “Point-Spread Function (PSF) Engineering and Adaptive Measurements for Single Molecule Super-Resolution Imaging”. This work is centered on enhancing the axial localization precision of fluorophore-tagged proteins in 3D using supercritical angle localization microscopy (SALM). For the said purpose, I have custom-built a Total Internal Reflection Fluorescence (TIRF) microscope capable of imaging cell membrane proteins. By accurately modeling the supercritical angle fluorescence into point spread function (PSF) of the microscope, all near field emitted fluorescence is detected into single channel. This approach achieves better localization precision (< 10 nm near coverslip) than conventional two-channel setups. Besides that, I have also contributed significantly to the development of MATLAB Instrument Control (MIC) and Single Molecule Imaging Toolbox Extraordinaire (SMITE), which are MATLAB-based software tools designed for instrument control and data analysis in single molecule fluorescence imaging. I am keen to apply my academic knowledge to contribute to the groundbreaking developments in fluorescence microscopy techniques. I am eager to leverage my comprehensive academic and practical experience in a challenging industrial role, aiming to make significant advancements in the development of new microscopy technologies. These advancements will have broad applications in biological research and healthcare, aligning with my long-term goals to impact these fields through development of advanced scientific tools and methodologies.
