RESEARCH1) Tumors escape immune surveillance by creating an abnormal state of tolerance towards their own antigens, thus escaping the body’s immune response; I have been examining IDO and the Kynurenine pathway and its effects on the growth and survival gliomas, meningiomas and metastatic brain tumors. 2) Study the distribution and the of effects of activated microglia, the central nervous system’s resident macrophage cells, inside the glioblastoma multiforme (GBM) tumor versus the infiltrating edge, and its effects on the tumor and microenvironment in terms of anti and pro inflammation and tumorigenesis properties. Also study the effect of osteopontin (OPN), overexpressed in a variety of cancers, and its role in tumorigenesis, inflammation, metastasis, and immune response. 3) Current projects in the lab involve metabolic reprogramming of glycolysis in GBM (both cell lines and primary neurosphere cultures) using simultaneous inhibition of dual metabolic pathways. We focus on lactate efflux using candidate small molecule inhibitors with concomitant targeting of mitochondrial dehydrogenase pathways.
