    • The genetics and molecular mechanism that regulate antidepressant SSRIs such as Prozac (fluoxetine) and Zoloft (sertraline) are widely prescribed for a variety of psychiatric conditions including depression, anxiety, and autism. These drugs inhibit the serotonin transporter SERT, which prevents the reuptake of serotonin. But the molecular mechanism of their action and their target are unknown. To further insight into the molecular mechanism of action of antidepressant, including the mechanism by which SSRI drugs are delivered to their molecular target, implementing a CRISPRi mediated genome-wide screen SSRI class drugs to identify targets as a gene required for the action of antidepressants drugs and validating their targets.     • Phm-2 lifespan extension in C. elegans through novel mechanism bacterial colonization, innate immune response and food avoidance behavior Studies of the microbiome are beginning to reveal the complex interactions between animal hosts and bacteria, but the effects on organismal aging remain poorly defined. we discovered that bacterial colonization can extend lifespan in the nematode C. elegans, a new and unexpected mechanism of delayed aging. We discovered a mutation that extends both health span and lifespan; the lesion affects the phm-2 gene, which encodes a protein homologous to human SAFB. Mutant worms have an abnormal pharynx grinder, which allows live bacteria to colonize the intestine. This defect shortens lifespan on highly pathogenic bacteria but extends lifespan on the standard E. coli diet by activating innate immunity pathways that lead to bacterial avoidance behavior and dietary restriction. • Angiotensin converting enzyme (ACE) inhibitor extends C. elegans life span To identify drugs that delay age-related degeneration and aging, screened several FDA-approved compounds and discovered that the novel drug Captopril and the acn-1 gene also caused a robust extension of lifespan and delayed age-related changes.• Identification of genes that regulate stress resistance and lifespan independent of Insulin signaling/FOXO by forward genetic screen. This research isolated two mutants by forward genetic screening that can cause lifespan extension independent of insulin/IGF-1 signaling.
