Thesis entitled ‘The Mechanisms of Opioid Tolerance’.My thesis focused on the involvement of protein kinase C (PKC) in the development and maintenance of opioid induced tolerance by looking at acute desensitisation, cellular tolerance, respiratory depression and locomotor activity. Interestingly, opioid tolerance has previously been shown to be reversed by low concentrations of ethanol and it has been postulated that PKC may play a role in this potentially lethal interaction.Throughout my PhD I developed expertise in a variety of fields including pharmacology, electrophysiology, behavioral techniques and neuroscience. Specific skills and techniques include: opioid receptor pharmacology, patch clamp electrophysiology, in vivo biology, whole animal plethysmography, locomotor activity, confocal microscopy, tissue culture and western blotting.
