As a project assistant in IGIB, I am involved in developing targeted assays using MRM-MS and also working on Metabolomics and data analysis of the same. I am also handling 5600+ Triple TOF and 6500+ QTRAP instruments (SCIEX) independently which has boosted my overall confidence in troubleshooting, method development, and experimental plans in OMICS.

A comprehensive metabolomics study with robust quality control procedures was performed using liquid chromatography-mass spectrometry (LC-MS) to characterize the urinary metabolic markers of different grades of UPJO and normal control subjects. The data were analyzed using the open-source web-based platform Workflow4Metabolomics (W4M). The metabolites in UPJO infants differed significantly from those of normal control subjects and were associated with the progression of UPJO. Metabolites involved in oxidative stress and renal disease pathways showed the most significant abundance differences.

Worked on the project entitled “Role of garlic extract in experimentally induced gastric cancer in male wistar rats”. Gastric cancer was experimentally induced by a nitroso compound MNU which was given to male Wistar rats for four weeks in drinking water followed by four weeks orally. This resulted in orthokeratotic hyperkeratosis leading to focal and atypical hyperplasia. But the administration of garlic extract resulted in the appearance of normal mucosa and gastric glands. This was also identified by analysis of the biochemical markers such as LDH, ALP, and gastrin. The possible mechanism of action of garlic extract in mitigating the lesions of the gastric cancer is by modulation of oxidation stress. This was confirmed by decreased lipid peroxidation and increased activity of the antioxidant enzymes.
