In vitro refolding of recombinant tryptophan synthase pathway enzymes were carried out by different methods such as refolding and by implying different expression conditions. Proteins were denatured using urea (8 M) as denaturing agents and refolding by gradual removal of denaturing agents. For proper refolding of tryptophan synthase α and tryptophan synthase β, the tryptophan synthase enzymes were His tagged at N-terminal and were co-expressed with the GroEL4S to get the soluble and enzymatically active proteins.
