  • I study enzyme-strubstrate interactions between the ClpXP protease and proteins targeted for degradation. I am specifically interested in substrates which require truncation and release by ClpXP in order to function.
  • I perform cloning, expression, and purification of recombinant proteins and chimeras, many of which have solubility or stability issues.
  • I assay the reactions via several methods, including coupled ATPase on a UV-Vis, direct observation of degradation on SDS-PAGE, and turnover of tagged fluorescent constrcuts.
  • I design and purify peptides mimicking ClpXP signal sequences and assay their binding affinity to ClpXP via fluorimetry or by probing peptide arrays with 35S-ClpXP.A secondary project is the study of a small peptide.
  • I design and purify peptide variants and test them against a library of purified ClpP homologs
  • I collaborate with another researcher to add a membrane-penetrating moiety to this peptide, which should enhance its in vivo activity.
