In this project, I investigated the engagement of anti-GRP78 autoantibodies to cell surface GRP78 on tissue factor activation, the primary pathway involved in clot formation. Identified and tested multiple small molecules to disrupt the interaction between cell surface GRP78 and anti-GRP78 autoantibodies using [techniques, e.g., high-throughput screening], contributing to potential therapeutic pathways in cardiovascular research.Trained and mentored incoming graduate students, fourth-year undergraduate thesis students, and summer research students.

Thesis Supervisor: Dr. Richard Austin (McMaster University, Faculty of Medicine)GRP78 is a protein chaperone primarily found in the endoplasmic reticulum. However, GRP78 has been found to localize to cell surface in certain disease states such as cancer and more recently atherosclerotic lesions. This project involves elucidating the role of cell surface GRP78 and identifying potential mechanisms of atherosclerotic lesion development. Using various molecular biology techniques such as RT-PCR, immunofluorescent staining, and Western Blotting, we demonstrated NF-κB pathway activation in response to GRP78 autoantibody binding. Results from experiments were used for publication.
