Working to provide deep phenotypic analysis of prospective and biobanked patient cohorts through the review of the latest available literature and studies. Working with collaborators to utilise this information to develop CRFs and biobank data collection plans. The QC of phenotypic data using Jupyter Notebooks to check cohort characterisation and the requested data point availability and distributions. Developing research questions based on the phenotype information and contributing to data analysis e.g., the Optimized Sequence Kernel Association Test (SKAT-O) analysis. The presentation of project progression and candidate gene selection to primary customers e.g., at AbbVie Joint Governance Committee (JGC) meetings.• Gene based analysis e.g., development, use and optimisation of the Optimized Sequence Kernel Association Test (SKAT-O) for AbbVie IBD candidate gene identification and Ionis NASH/NAFL candidate gene support. Development of notebooks for post-run analysis.o Replication of candidate gene significance in other datasetso Candidate gene robustness across different iterations of SKAT analysiso Analysis of variant distributions driving SKAT-O signal • Literature review of candidate genes as potential disease therapy targets, including:o Tissue specific expression of candidate gene transcriptso Biological pathways and molecular function of gene encoded proteinso Variant interpretation; annotation of variants for max consequence, gene variant tolerance and protein sequence conservation• Writing integrated GOR/NOR & R code to provide tools for data analysis in Sequence Miner and Jupyter Notebooks• CRS for the following cohorts: o Diabetic Nephropathyo Parkinson’s Diseaseo Brain Tumourso Multiple Sclerosis• Coordination with collaborators to collect, QC and harmonise phenotypic data• Regular presentation of data at customer technical and JGC meetings• Health and Safety Representative for the Research Team

Working in Dr Rob van Montfort’s Hit Discovery and Structural Design team in collaboration with Astex Pharmaceuticals on a drug discovery project targeting MMSET (NSD2) a histone methlytransferase whose overexpression is associated with poor prognosis in multiple myeloma patients.• Use of molecular and cell-based techniques to develop assays measuring the ability of small molecule compounds to inhibit the target protein and to study the mechanism of action in cells.• Review of the latest literature to better understand the role and regulation of the target gene and the clinical implications for its inhibition. • Presentation of data at joint ICR and Astex Pharmaceuticals project meetings

Working in Professor Richard Marais’ Signal Transduction Team on a Wellcome Trust funded drug discovery project. The project aimed to develop novel therapeutic agents for the treatment of metastatic cancers by targeting the activity of lysyl oxidase (LOX): An enzyme active in the extracellular matrix and shown to play a critical role in metastasis.• Developed and ran an array of biochemical and cell-based assays to test the efficacy of targeted inhibitors.
