With mouse models of Wiskott-Aldrich syndrome and X-linked neutropenia, we investigated the effect of functional mutations in the Wiskott-Aldrich syndrome protein on dendritic cell adhesion and migration. Cell adhesion was measured in primary mouse cell cultures using confocal microscopy followed by ImageJ analysis with custom macros. Changes in dendritic cell migration in vivo were evaluated by a flow cytometry-based cell migration assay. Understanding these pivotal aspects of dendritic cell function may provide insight into immune regulation and aid immunotherapy design.

Using primary mouse cell cultures and neuroinflammatory mouse models we identified compounds capable of mitigating microglia activation. The immunomodulatory effects were characterized in vitro with RT-PCR, ELISA, and cytometric bead arrays, while the in vivo effects were observed with animal behavior studies. These findings demonstrate the high plasticity of microglia and the potential of repurposing compounds for the management of microglia activation under neuroinflammatory conditions.

The lysine demethylase KDM4B contributes to IL-8 production via epigenetic regulation. ELISA and RT-PCR revealed that KDM4B inhibitors do indeed reduce IL-8 expression in a gastric cancer cell line co-cultured with H. pylori, highlighting their potential as a treatment option for gastric and other cancers.
