    Worked to monitor the tau-tubulin interactions in live neurons in culture and in vivo using Second Harmonic Generation. Utilizing various molecular, microscopic, cell biological, and biochemical techniques to understand the microtubule network and how it's influenced by tau. Using SHG and microfluidic devices, I was able to show a difference between 3 and 4 repeat tau in living neurons and how they interact with the microtubule network in the axon of neurons in culture. I was also able to demonstrate how certain tau mutations found in tauopathies can affect the interaction between tau and the microtubule cytoskeleton and actually make tau bind tighter to the microtubule.

    During my graduate student experience I gained experience in a wide variety of techniques. Initially I had experience in making and culturing rat hippocampal slice cultures to be used for patch-clamp experiments. The majority of my work in grad school focused on cell cutlure models utilizing mammalian cell lines to model tau phosphorylation events in various toxic and insulting conditions and how the Wnt signaling pathway could influence that phosphorylation. Some basic techniques included cell culture in sterile envirnoment, gene cloning, and transfection, Western blotting, immunocytochemsitry, autoradiography, ex vivo assays for kinase and phosphatase activities. Also gained extensive experience in generating and characterizing multiple stable cell lines with various selection cassettes.
