Managed and led NGS studies to deliver novel genetic/genomic knowledge in multiple preclinical species.Drove high dimensional omics studies to investigate MOA of high value targets, identified clinical biomarkers, contribute to patent and IND filing.
Sr. Clinical Research Associate
Responsible for bioinformatics pipeline building, advanced statistical analysis, machine learning algorithms, customized high-throughput experimental design and predictive modeling of complex diseases. Identified common and rare variants associated with Age-related macular degeneration (AMD).• Designed and analyzed large-scale (3200 samples) targeted NGS sequencing study and identified novel rare functional variants in C3, C9 and CFI causing AMD. • Investigated rare variants for AMD using GWAS and Gene-based tests on large samples (10,000) genotyped by custom-designed Illumina whole-exome array. • Maintain and support the BWA-GATK pipeline, manage large NGS dataset on linux cluster. • Designed and Implemented whole-exome sequencing study in selected AMD families and identified new rare functional variants causing AMD. • Led large-scale meta-analysis of Genome-wide association study (GWAS) for AMD. Identified two novel loci and confirmed the findings in 10 replication cohorts of more than 60,000 samples. • Established risk assessment tools that predict AMD progression based on genetic and environmental factors. Licensed to Arctic Diagnostics in Macular Risk® kit for commercial genetic testing.• Participated in the international AMDGENE consortium and played key role in the meta-analysis of GWAS, pathway analysis, eQTL analysis, interaction and epistasis analysis. • Led candidate gene study for genetic variants associated with risk of choroidal neovascularization (CNV) events in the high myopic patients.
Collaborator
Postdoc Fellow
• Investigated genetic association to AMD in a GWAS dataset of discordant sibpairs and families.• Studied genetic association to opiod dependence in a substance dependence GWAS cohort.
Phd Candidate In Bioinformatics
• Developed novel analytical tools based on the random forests algorithm that can be applied on dataset of families and sib-pairs, and robust to population admixture. • Mapped multiple QTLs for cholesterol levels by quantitative traits linkage analysis in large, consanguineous families with atherogenic dyslipidemia. (In collaboration with GlaxoSmithKline)• Identified two nicotine dependence genes (ANKK1 and DDC) in a family-based association study.• Associated the MANEA gene with cocaine related behavior in families of substance dependence. • Evaluated methods of linkage studies using SNPs and STR markers in the dataset of Genetic Analysis Workshops (GAW14).• Constructed a MySQL database and web-browser interface for the cell signaling pathways study to integrate experimental data from B-cell ligand screen, yeast two-hybrid and microarray.
