Short-read sequencing has notable drawbacks for studying repetitive areas of the genome. As such, researchers are actively seeking means to contiguously sequence longer strands of DNA. Currently researchers can use long-range PCR and large hybridization capture, but both technologies have a limited tolerance for random structural variance, making capture of these regions challenging. Enter Cas12a, which has unique properties that allow it to cleave at designated sites despite unknowable structural variation, resulting in predictable capture of DNA from regions of interests. TTT's purpose was to harness the unique capacities of Cas12a to produce long read library preparation kits for Illumina, Oxford Nanopore, and PacBio, designed for researchers studying long-non-coding portions of the Genome; repeat regions high in GC content; and structural variations such as insertions, deletions, and repeat expansions.
