In this role, I supported CRF's Swim Guide water quality monitoring program to provide free and accessible water quality information for a myriad of testing sites chosen based on their popularity as swimming holes and proximity to known sources of pollution. My main duties involved coordinating with volunteers and fellow interns to collect water samples and associated site data. Each week, the interns at our location performed E. Coli tests and water quality tests for all sites in our section of the basin and published the data on the swim guide websites and CRF social media. In addition to my main duties, I was also able to assist in our Kayak operations, educational programs, Eco-Tours and miscellaneous maintenance tasks at our Boathouse Location. This position allowed me to develop a more robust sense of professionalism under the guidance of senior CRF staff as well as numerous invaluable professional relationships for which I am continually grateful.

My team conduced a baseline survey of antimicrobial resistance genes in environmental bacterial samples in the Catawba River Basin. Data collection involved collecting water samples from several public river access points spanning the Charlotte-Metropolitan area over a period of three months, with each location chosen for its proximity to point source polluters. Bacterial cultures were isolated from the samples. A second sample from each location underwent laboratory testing for a variety of water quality parameters. Our initial testing of our bacteria involved incubation after application of a range of broad spectrum antibiotics to track susceptibility to these compounds. The samples then underwent PCR amplification and Agarose gel electrophoresis to allow for Sanger sequencing. Sequence data was used for the identification of our bacteria and tracking of our chosen genetic markers. Our next phase involved comparing the susceptibility of our samples to the Comprehensive Antibiotic Resistance Database. The final phase of our project would have included analysis for the presence of genes with a known role in conferring resistance. Unfortunately, we were unable to complete this phase due to Covid-19 related shutdowns. This position provided me with essential laboratory skills which I hope to develop further in future research roles.
