At A2 Biotherapeutics I am part of a collaborative research team developing next-generation cell therapies for cancer. My focus is investigating the biophysical mechanisms of CAR-T cell function, and developing novel engineering strategies to enhance and broaden CAR-T functionality. A2 has developed a logic-gated CAR-T platform integrating a safety switch to address toxicity, known as Tmod, and my work aims to expand the Tmod platform to a broader patient population.

As part of the Integrative Design team at Amgen, I led purification and analytics efforts for novel antibody-based multispecific modalities. I also co-developed protein engineering projects focusing on evaluating innovative therapeutic platforms, facilitating bispecific production, enabling high-throughput bispecific evaluation, and building in silico methods for predicting bispecific properties.

Mentor: Yunsun Nam, PhDGraduate Program: Molecular BiophysicsDuring my graduate training, I investigated the structural and biophysical basis for how human primary microRNAs are recognized and processed. This work led to major insights into the RNA/protein interactions that underlie the recognition of diverse microRNA substrates, and how this process is regulated. During this time I pioneered numerous in vitro RNA/protein assays and applied a broad set of molecular tools to generate, interpret and validate the first Drosha/DGCR8/pri-miRNA structures.

Research Mentor: Jonathan Ploski, PhDI developed a workflow for using laser-capture microdissection to isolate sub-regions of rat brain slices for gene expression analysis. Using this strategy I evaluated quantitative differences in RNA expression patterns between adjacent (but functionally distinct) subnuclei of the amygdala. I also established a protocol for in situ hybridization of rat brain tissue and applied this method to validate my RNA expression data. I also worked to develop a system for using high frequency electrical stimulation to alter gene expression in the rat hippocampus, and pioneered a method for isolating distinct layers of the hippocampus to evaluate stimulation-induced gene expression changes.

I worked as a tutor/instructor for an advanced biochemistry course of 150+ students. I held weekly group discussions reviewing course content, which rigorously covered the pathways involved in the biosynthesis and breakdown of amino acids, nucleotides and lipids.

Mentor: Lucien Tres Thompson, PhDI co-led a project investigating the behavioral and neurophysiological effects of a small molecule proposed to be a potential neurotherapeutic. I coordinated and oversaw behavioral assays examining the effects of this compound on spatial and aversive memory in the rat brain. I also developed detailed protocols for precisely isolating neuroanatomical structures from the rodent brain using a cryostat microtome, prepared live rodent brain sections for electrophysiological experiments, and trained 20+ students in laboratory methods for neuroscience.

I volunteered as a teaching assistant for a senior-level neurophysiology course covering ion channel function and the dynamics of synaptic transmission. I created and led an online discussion forum for students to ask/discuss questions and share additional online resources. I also held weekly review sessions, met one-on-one with students for additional tutoring, created original course material and assisted in proctoring exams.
