I found, using localized microdialysis, that Neostigmine Bromide, a cholinesterase inhibitor, increased GABA levels and decreased EEG delta power in the oral part of the pontine reticular formation of B6 mice. I also observed the same results following systemic Physostigmine injections (i.p) in B6 mice. I then conducted experiments to assess the mechanism of action of both these cholinesterase inhibitors that included co-administration of scopolamine, a muscarinic antagonist, and bicuculline, a GABA receptor competitive agonist: scopolamine blocked the increase in GABA levels and bicuculine blocked the decrease in EEG delta power.
