The Orr lab studies genes that play a role in neurodegeneration, specifically focused on the molecular basis of the disease spinocerebellar ataxia type 1 (SCA1). SCA1 is a dominantly inherited neurodegenerative disease that is caused by an expanded polyglutamine repeat in the protein ataxin-1.Under the guidance of Dr. Michael Andresen I learned a variety of cell culture and biochemical techniques:• Propagated stably transfected cell lines, performed transfections, and learned basic cell culture techniques• Performed site-directed mutagenesis of ataxin-1 and purified proteins through affinity column chromatography • Analyzed biochemical interactions of mutated proteins in GST pull-down assays• Used size-exclusion chromatography to study ataxin-1 protein complex• Developed a site specific phosphorylation assay to screen for potential kinases which phosphorylate ataxin-1

•I worked with Sarah McAvoy (Ph.D.) on a project studying common fragile site breakage. Common fragile sites are regions of extreme genomic instability found in all individuals. Contained within many of these fragile sites are very large genes, which have been implicated to play a role in cancer development.•We performed FISH to fluorescently label fragile sites and scored breaks on human chromosomes.
