1. A genetic and clinical study of Osteopathia Striata with Cranial Sclerosis. Osteopathia striata with cranial sclerosis (OSCS) is an X-linked dominant sclerosing bone dysplasia that presents in females with macrocephaly, cleft palate, mild learning disabilities, sclerosis of the long bones and skull, and longitudinal striations visible on radiographs of the long bones, pelvis, and scapulae. In males this entity is usually associated with fetal or neonatal lethality. The study of a large set of patients, all diagnosed with OSCS, expanded the genetic and phenotypical spectrum and elucidated possible correlations between the genetic aberration and the clinical consequences. The overview of all mutations and their associated clinical features provides a valuable resource for both the molecular and clinical geneticist dealing with individuals with OSCS. 2. The study of two well-known natural mutants of the rat characterized by osteopetrosis, the ia and the op rat.The osteopetroses are a genetically heterogeneous group of bone disorders characterized by a reduction in bone resorption resulting in an overall increase in skeletal mass. In human, different types of osteopetrosis can be distinguished based on the severity, the mode of inheritance and the age of onset. In animals, at least 20 models of osteopetrosis -both induced and spontaneous mutants- have been described. The general aim of the project was to gain novel insights into the bone resorption process and the mechanisms by which this is performed by the osteoclasts. We tried to achieve this by studying two well-known natural mutants of the rat characterized by osteopetrosis, the incisors absent (ia) and the osteopetrotic (op) rat.
