My role in Ribometrix is to validate targets for small molecule screening by studying the complexity and functionality of their RNA structure.- Set up of new lab space (BSL2) and acquisition of supplies and equipment- Continued development of techniques to study RNA structure and subsequent impact on health and disease- Validation of targetable RNA structures for small molecule screening using SHAPE-MaP with MiSeq and NextSeq platforms - Development of cell-based functional assays for target and hit validation for read-out in a plate-reader. Includes luciferase and fluorescence assays.- Health and Safety: Development of robust Lab Safety Plan and training regimen- Coordination of team building activities

• Prepared transcription-based GFP reporter strains and used fluorescence assays to study the role of temperature, pH, rovA, H-NS, and psaEF on the expression of virulence factor psaA in Y. pestis. Also investigated the role of myfEF in the regulation of myfA (homologs to psaEF and psaA, respectively) in Y. enterocolitica (Manuscript in progress).• Prepared samples for histology and used immunohistochemistry to study the role of Y. enterocolitica toxin YaxAB in a mouse model of oral infection (Wagner et al, 2013).

• Examined the mechanism by which RBM9 and hnRNP H1 regulate FGFR2 exon IIIc splicing by construction and use of stable cell lines with fluorescent splicing reporters and nuclear extracts (see Mauger et al, 2008 in Publications below) • Began development of a genome-wide shRNA screen to identify regulators of FGFR2 alternative splicing and factors associated with epithelial to mesenchymal transitions.

• Cloned small RNAs from tissues, cultured cells, and bacteria to form small RNA libraries, including piRNAs from human testis tissue samples (Landgraf et al, 2007)• Tested the activity and specificity of the catalytically active T4 RNA ligase 2 (1-249) K227Q that was under development in the laboratory as a tool for microRNA cloning, now available through NEB (Hafner et al, 2008).• Characterized polyclonal antibodies for specificity to Ago1, Ago2, Ago3, Ago4, Hiwi1, Hiwi2, Hiwi3 for use in immunoblotting, and monoclonal antibodies specific to Ago2 for detection by ELISA, immunoprecipitation, and immunoblotting.
