The team is currently working on developing and validating anti-HIV-1 cell therapies using NK cells as well as Dual-affinity re-targeting (DART) antibodies. We are looking to deepen our understanding of NK functions, especially during HIV infection, and of how to harness these in order to fight HIV. We are trying to develop NK cells (transduced or activated and expanded under specific conditions) to use as potential treatment for HIV. For that purpose we are developing in parallel a mouse model (in NSG) to verify our findings.Using: Work in BSL-2+, Cell Culture of established cell lines and primary cells from blood (healthy and infected subjects), immuno-magnetic cell sorting, cell transduction with retrovirus, BSL-2 animal handling, Mouse injection (IP, IV, SubCu, IM) and bleeding (tail nick, sub-mandibular, cardiac puncture), kit ELISAs, Virus and Cell Assays, Flow Cytometry (Attune), Muse cell analyzer

The Laboratory for Tumor and AIDS Virus Research, Principal investigator: Dr. Jay A. Levy, M.D.Department of Medicine, Division of Hematology and OncologyThe team is focusing on finding, validating, and developing anti-HIV-1, and, more broadly anti-viral proteins. We are looking into the CD8+ cell non-cytotoxic control of HIV-1, and the precise pathways activated in this condition. Dr. Levy and I successfully wrote and submitted an R21 grant with the NIH, which we were awarded. We are also involved in a stem cell project, in which the team is developing a way to mutate patients iPS cells to make them resistant to HIV-1 infection. The goal being to safely re-introduce the mutated iPS cells back into patient for treatment and possibly a cure. Mice study with human iPS cells are on going.Using: Cell Culture of established cell lines and primary cells from blood, immuno-magnetic cell sorting, cell differentiation, cell transfection and electroporation, bacterial transfection, MaxiPreps, kit ELISAs and in house ELISAs, Virus Assay, Radioactive and Colorimetric RT Assay, Flow Cytometry (LSRII, LEGENDplex, FlowJo), SDS-PAGE, Western Blot, Native Gels, Protein and Nucleic acid extraction, RT-PCR, q-PCR, PCR, sequencing, bioinformatics, protein immunoprecipitation.
