Bode, where previously a novel potassium acyltrifluoroborate (KAT) - hydroxylamine ligation reaction showed potential but had not been applied as a method for bioconjugation. We realized this potential by incorporating hydroxylamines onto the surface of recombinant proteins and reacted them with various macromolecular KAT reagents. Optimization of the ligation conditions enabled not only the site-specific PEGylation but also the dimerization of complex proteins in a stoichiometric fashion.

The site-specific integration of linear fragments into macrocyclic entities from a purely synthetic approach was successfully demonstrated for the first time. Finally, a robust and efficient multi-step synthesis of coveted aziridine aldehyde dimers was developed.
