Senior Research Associate Ii
Conducted cutting edge research, providing experimental support in conjunction with those working to model protein motion and small molecule interactions in silico. Provided lab operations management to support company goals while also conducting research at the bench. • First bench biologist hired when company was in stealth mode (employee number 8). Responsible for the initial set-up of the biology lab space; populating the empty lab with all equipment and supplies required to carry out cutting edge research • Conducted research on oncology targets that progressed to the clinic (SHP2, PI3K and FGFR2), among others • On the team for the design and implementation of entirely new lab and office build-out when it was time to expand the company in 2018. Involved setting up space for tissue culture, bench work, chemistry needs, equipment infrastructure, freezer and chemical storage space as well as office and meeting space • As company continued to grow, provided input on equipment and workflow needs of a second building expansion space in 2021 • Vetted and implemented two ELN systems (Arxspan, then LabGuru). Trained new hires in the use of ELN and associated cell inventory system • Maintained cell bank with periodic testing for mycoplasma and validation of cell lines via STR • Acted as point person for lab equipment service calls and service contracts. Triaged requests, tracked PMs and determined the value-added nature of service contracts from vendors based on knowledge of equipment reliability and maintenance needs • Managed lab consumables, tracked usage (using Sortly inventory software) and worked with vendors to negotiate pricing discounts • Assisted in the setup of lab equipment monitoring systems (Tetrascience, Elemental Machines). Determined what equipment needed monitoring, what thresholds to track and training of lab personnel in response to alerts
Research Associate
Independently conduct research on targets related to the USP7 pathway. • Assay development and implementation to elucidate cellular processes that determine the target specific effectiveness of compounds • Assist Biology Group with automation needs, and transfer technology to increase efficiency and robustness of data • Collaborate with the high throughput screening group to move projects forward from the benchtop to the HTS platformProvide automation assistance to colleagues, writing and validating protocols to increase throughput, accuracy, precision and consistency of experiments.
Scientist 2
Independently develop, manage, and coordinate transfer of high-throughput screening assays and platforms for oncology projects. • Received a “Catalyst Award” from Novartis for work on a screening assay that was later implemented widely across the multiple sites at Novartis • Coordinated the implementation and maintenance of multipurpose automated screening platforms • Conferred with vendors to design, purchase, build and test new systems, while continuing to maintain and repurpose diverse equipment for changing needs • Assessed workflows and consumables for assays • Developed cell-based assays for shRNA target validation and transfer technology to fellow researchers • Developed assays to be utilized by other project groups, testing multiple compounds in various combination grids on large panels of cell lines • Transferred technology to researchers through training and support and document SOP’s for new assays and equipment use and maintenance
Senior Research Associate
Independently implemented novel solutions to improve research methodologies.· Imaged cells with fluorescent dyes and fluorescently labeled IHC for fluorescent and confocal microscopy (Metamorph system) to assess phenotypic and morphological characteristics in response to treatment with oncology related compounds.· Developed a 3D cell culture system on an artificial extra-cellular matrix (ECM) to approximate an in vivo phenotype and response for compound screening.· Profiled the protein expression (in response to treatment) of signal transduction pathways in breast and non-small-lung cancer cells lines grown on ECM and plastic by Western blot. Based on data obtained, designed experiments to elucidate apoptotic pathways.· Conducted cell-based assays on oncology combination trial-candidates, using plate-based cell proliferation/metabolism assays (WST, CellTiterGlo), cell death assays (ApoOne, Cytotox-One), and Flow Cytometry (FACS, Guava PC). - Collaborated in the design and implementation of an automated imaging and picking system for high throughput mammalian cell line development. The automated system enabled the detection and picking of high expresser clones from colonies.
Research Associate
Conducted experiments with minimal supervision and aided coworkers in rest of the company with the implementation of new technology and scientific methodology. · Designed and tested a novel, small volume, disposable airlift reactor system to determine conditions needed for scale up of cultured mammalian cells for optimal protein production in a bioreactor. Produced a working system with temperature, pH, gas and nutrient control. This allowed the maintenance of several independent cultures with sufficient volume to sample from, utilizing a bench top footprint. · Developed a cell based screening assay for a library of 50,000 GPCRs, utilizing modified amphibian cells (melanophores). This involved the programming and maintenance of an in-house customized robotic platform for spotting of compounds and imaging of cell response, along with all of the cell culture involved in the assay.· Modification of BAC (Bacterial Artificial Chromosome) vectors. Assisted in the subcloning and mutagenesis of the BACs and supervised contract workers in the high throughput transformation, culture and DNA preps from bacterial cultures. Using this technique we were able to discover novel full-length gene transcripts. · Validated a modified plasmid prep method in collaboration with the LigoChem Corp. Automated the protocol for a robotic high-throughput system. · Introduced and validated automated systems to better advance the performance of various groups within the company. · Troubleshot and conducted onsite maintenance and programming of automated systems (Rapid Plate, PlateMate, MotoMan), and coordinated service from vendors when needed. · Assisted in experiments using standard molecular biology techniques, including sequencing, recombinant cloning, mutagenesis, transformation and transfection.
