Independently implemented novel solutions to improve research methodologies.· Imaged cells with fluorescent dyes and fluorescently labeled IHC for fluorescent and confocal microscopy (Metamorph system) to assess phenotypic and morphological characteristics in response to treatment with oncology related compounds.· Developed a 3D cell culture system on an artificial extra-cellular matrix (ECM) to approximate an in vivo phenotype and response for compound screening.· Profiled the protein expression (in response to treatment) of signal transduction pathways in breast and non-small-lung cancer cells lines grown on ECM and plastic by Western blot. Based on data obtained, designed experiments to elucidate apoptotic pathways.· Conducted cell-based assays on oncology combination trial-candidates, using plate-based cell proliferation/metabolism assays (WST, CellTiterGlo), cell death assays (ApoOne, Cytotox-One), and Flow Cytometry (FACS, Guava PC). - Collaborated in the design and implementation of an automated imaging and picking system for high throughput mammalian cell line development. The automated system enabled the detection and picking of high expresser clones from colonies.

Conducted experiments with minimal supervision and aided coworkers in rest of the company with the implementation of new technology and scientific methodology. · Designed and tested a novel, small volume, disposable airlift reactor system to determine conditions needed for scale up of cultured mammalian cells for optimal protein production in a bioreactor. Produced a working system with temperature, pH, gas and nutrient control. This allowed the maintenance of several independent cultures with sufficient volume to sample from, utilizing a bench top footprint. · Developed a cell based screening assay for a library of 50,000 GPCRs, utilizing modified amphibian cells (melanophores). This involved the programming and maintenance of an in-house customized robotic platform for spotting of compounds and imaging of cell response, along with all of the cell culture involved in the assay.· Modification of BAC (Bacterial Artificial Chromosome) vectors. Assisted in the subcloning and mutagenesis of the BACs and supervised contract workers in the high throughput transformation, culture and DNA preps from bacterial cultures. Using this technique we were able to discover novel full-length gene transcripts. · Validated a modified plasmid prep method in collaboration with the LigoChem Corp. Automated the protocol for a robotic high-throughput system. · Introduced and validated automated systems to better advance the performance of various groups within the company. · Troubleshot and conducted onsite maintenance and programming of automated systems (Rapid Plate, PlateMate, MotoMan), and coordinated service from vendors when needed. · Assisted in experiments using standard molecular biology techniques, including sequencing, recombinant cloning, mutagenesis, transformation and transfection.
