New technologies allow us to better understand the world around us and the world inside us.Over the last 10 years, I have been building high resolution microscopes🔬(nanoscopes!) in collaboration with biologists 🧫, developing and integrating advanced hardware and software technologies into simple user workflows that allow us to better understand the molecular mechanisms of diseases and how to treat them. Included optimization of User Interface, Multithreading, and the implementation of several algorithms to improve single molecule localization in 3D, on multiple cameras, and in high-density acquisitions.- Co-coordinated microscope updates to enable dual-objective SMLM in a "user-friendly" configuration, including mechanical/stability improvements.- Co-designed optical unit enabling spectral SMLM in a dual-objective configuration.- Participated in numerous collaborations, conferences, and interdisciplinary projects, most notably for the detection, quantification and tracking of membrane receptors in neuron cell cultures.

As a CIFRE PhD Student, I worked in collaboration between the company Imagine Optic and a research lab at the Interdisciplinary Institute for Neuroscience on Adaptive Optics in Single Molecule Localization Microscopy applications. • Developed and deployed a software plugin integrating Imagine Optic's MicAO adaptive optics software into the MetaMorph acquisition environment, facilitating the use of adaptive optics for untrained microscopists.• Cultured, fixed and labelled a variety of biological samples, ranging from cultured COS and HeLa cells to organotypic brain slices.• Utilized various single molecule localization -based "super-resolution" microscopy methods, from single particle tracking (sptPALM, uPAINT, QD-Tracking) to long-scale imaging of fixed samples (PALM, dSTORM, DNA-PAINT).• Implemented MicAO adaptive optics system on an multiple existing semi-commercial single molecule localization microscopes. Demonstrated the advantages of AO for astigmatism-based 3D single molecule imaging in cell cultures and organotypic brain slices.• Developed a new single molecule microscope capable of spectral detection without compromising spatiotemporal resolution, for use with cultured cells.• Developed new and improved existing internal single molecule localization software. Improvements include multithreading, visualization and UI/UX improvements. New developments included the algorithms and field of view transformations required for quantitative spectral imaging using a dual-objective approach to image the same focal plane (VB.NET / C#).• Developed the strong communication skills required to thrive in an extremely interdisciplinary environment

Project: Determination of AMPA Receptor Subunit Composition using dSTORM Superresolution Imaging. Performed cell culture and antibody labeling of heterologous cells. Imaged samples using dSTORM super-resolution microscope. Created new visualization technique using MATLAB, resolving features smaller than the super-resolution pixel size.
