As project leader for a potential spin-off project, I was accompanied by several of the main players in the French DeepTech startup ecosystem (AST, CNRS RISE, UNITEC) in an effort to commercialize an innovative 3D microscopy technique.After several years of entrepreneurship training and lots of customer development, the final incarnation of REFLECT planned to make 3D cell cultures fast and easy to use, helping biologists and biotech companies understand how their cultures are growing and saving them time and money by stopping unsuccessful cultures early.I ultimately decided to put the project on hold until the 3D cell culture market matures a bit -- But I'm still a huge fan of 3D cultures + 3D imaging :)

Team: Quantitative Imaging of the cellAdvisor: Jean-Baptiste SibaritaProjects: - Adaptive Optics for Single Molecule Superresolution Microscopy,- Dual objective, spectrally-displaced SMLM for enabling multicolor single particle tracking of ≥3 proteins.Skills:- Led the development of PALMTracer version 2, a single molecule localization software plugin for the Metamorph image analysis software (VB.NET / C#). Included optimization of User Interface, Multithreading, and the implementation of several algorithms to improve single molecule localization in 3D, on multiple cameras, and in high-density acquisitions.- Co-coordinated microscope updates to enable dual-objective SMLM in a "user-friendly" configuration, including mechanical/stability improvements.- Co-designed optical unit enabling spectral SMLM in a dual-objective configuration.- Participated in numerous collaborations, conferences, and interdisciplinary projects, most notably for the detection, quantification and tracking of membrane receptors in neuron cell cultures.

As a CIFRE PhD Student, I worked in collaboration between the company Imagine Optic and a research lab at the Interdisciplinary Institute for Neuroscience on Adaptive Optics in Single Molecule Localization Microscopy applications. • Developed and deployed a software plugin integrating Imagine Optic's MicAO adaptive optics software into the MetaMorph acquisition environment, facilitating the use of adaptive optics for untrained microscopists.• Cultured, fixed and labelled a variety of biological samples, ranging from cultured COS and HeLa cells to organotypic brain slices.• Utilized various single molecule localization -based "super-resolution" microscopy methods, from single particle tracking (sptPALM, uPAINT, QD-Tracking) to long-scale imaging of fixed samples (PALM, dSTORM, DNA-PAINT).• Implemented MicAO adaptive optics system on an multiple existing semi-commercial single molecule localization microscopes. Demonstrated the advantages of AO for astigmatism-based 3D single molecule imaging in cell cultures and organotypic brain slices.• Developed a new single molecule microscope capable of spectral detection without compromising spatiotemporal resolution, for use with cultured cells.• Developed new and improved existing internal single molecule localization software. Improvements include multithreading, visualization and UI/UX improvements. New developments included the algorithms and field of view transformations required for quantitative spectral imaging using a dual-objective approach to image the same focal plane (VB.NET / C#).• Developed the strong communication skills required to thrive in an extremely interdisciplinary environment

Project: Determination of AMPA Receptor Subunit Composition using dSTORM Superresolution Imaging. Performed cell culture and antibody labeling of heterologous cells. Imaged samples using dSTORM super-resolution microscope. Created new visualization technique using MATLAB, resolving features smaller than the super-resolution pixel size.

Implemented a 4f-imaging based beam-steering system. Obtained spectroscopic information of trapped objects in back-reflection. Demonstrated trapping of latex beads and live yeast cells. Developed strong skills in laser and optical system alignment and function.
