PI: Jun J Yang; Pharmacotyping Core, Pharmaceutical Sciences Department• Generated correlation heat maps comparing the drug response across more than 500 unique samples and a 14-drug panel in RStudio by developing functions using “tidyverse” libraries to clean and organize the data and “reshape2” and “ggplot2” libraries to analyze the correlation and visualize the heatmap.• Developed a set of functions in RStudio to examine the individual drug response histograms, drug-drug scatter plots with loess curve, and drug-drug correlation and significance levels for a 14-drug panel and visualized the results in a single figure.• Examined the relationship between drug response of our 14-drug panel and the subtype of each ALL patient sample.• Oversaw and coordinated the purchase and installation of the PerkinElmer Operetta CLS High Content Imaging System as well as incubators and Biological Safety Cabinets for the new room for the Pharmacotyping core.• Coordinated training on the new instrument with Field Application Scientists from PerkinElmer and team members with various imaging projects.• Worked with the Pharmacotyping team to develop a high content imaging assay to increase our drug panel from 16 to 40 drugs and reduce the number of cells used by 60%.• Acquired a liquid handling system and designed custom functions to decrease plating times at every stage in the process.• Created Standard Operating Procedures and QC measures to ensure reproducibility of the imaging assay and reliability of all results.• Received requests from collaborators and designed experiments to examine drug sensitivity of samples and cell lines of interest and/or newly developed drug molecules.

Dr. William Evans Lab, Pharmaceutical Sciences Department• As a technologist in the Pharmacogenomics Lab of Dr. Evans, I contribute to the study of the genetic determinants and mechanisms of drug resistance in Pediatric and Adult Acute Lymphoblastic Leukemia (ALL) as part of the Center for Precision Medicine in Leukemia (CPML) Project• Lead technician for CPML functions: coordinate the timely receipt of fresh patient samples through effective communication with our internal biorepository and external collaborators; process newly arrived samples by determining viability, red blood cell (RBC) contamination, and blast percentage via staining procedures and microscopic inspection; improve sample quality through RBC Lysis and MACS Blast enrichment protocols to meet pre-testing quality standards; distribute cells to Postdoctoral fellows and collaborating labs within the institution• Utilize cell culture techniques to perform Bioassays to determine drug sensitivity in Primary patient samples via an MTT-based absorbance assay for cytotoxic agents and a flow cytometric assay for targeted agents; Extraction of DNA and RNA from tumor samples for genetic sequencing• Keep meticulous records for every sample received and update corresponding records in the project database; Prepare weekly presentations summarizing the samples received and testing performed; Utilize Prism software to create drug summaries and analyze pilot and cell line experiments; Update Standard Operating Procedures as necessary to ensure the continued quality and repeatability of all aspects of the project• Identified and addressed QC issues such as data artifacts and increased background signal in the MTT bioassay through the utilization of cell line experiments• Assist postdoctoral fellows and Ph.D. students in designing and conducting their experiments; Ensure that reagents and lab supplies are stocked and within expiration, as Preparation of reagents for collaborators
