    I worked in collaboration with CRUK-TDL on a variety of projects such as:• Validation of novel immune-oncology targets.• Inhibition of novel immunometabolism targets.• In silico quantification of tissue fibrosis.To facilitate this research, I cultured cells to perform siRNA knockdown and generate cytoblocks as positive and negative controls. I utilised immunohistochemistry techniques to conduct antibody validation. Once an antibody has been validated, I developed multiplex immunofluorescence assays and performed staining across a variety of normal and cancer tissues including: breast, colon, heart, lung, pancreas, prostate, skin and thymus.After staining, I scanned and analysed the slides using multiple analysis platforms including inForm, QuPath and VisioPharm.I presented my research at monthly intervals to keep the team at Leicester Cancer Research Centre and CRUK-TDL updated on my current project. To aid in the understanding of the results I also created video tutorials explaining tissue specific histopathology in relation to the target of interest.

    My project focused on screening novel anti-trypanosomal 6-benzimidazol-1-ylbenzothiophenes against A2780 ovarian carcinoma cells as a secondary toxicity test of previously identified hit compounds. Techniques and skills: SNAr reactions, cyclisation reactions, thin layer and chromatography, fractionation, A2780 ovarian carcinoma cell culture, seeded cells in a 96-well format, applied drug dosing regimens, conducted MTT assays, maintained a laboratory book, calculated and applied appropriate laboratory calculations, wrote up materials and methods for publication. Based on these results I designed 3 potential analogues inspired by state-of-the-art research, which conformed to Lipinski’s rule of 5. Of the compounds designed from the study, the benzoxaborole derivate was the most pertinent as its low molecular weight and lipophilicity exhibit properties desired for crossing the Blood Brain Barrier.5000 word write up of the project, entitled: Cytotoxicity screen of hit novel antitrypanosomal 6-benzimidazol-1-ylbenzothiophenes received 76%.

    Research project: Synthesis and Biological Evaluation of Novel Antiprotozoal Drug Scaffolds.I was the first student at De Montfort University to be awarded the Wellcome Trust Biomedical Vacation Scholarship during my second-year summer vacation. During the scholarship I synthesised 10 rationally designed analogues via SNAr and cyclisation reactions, and elucidated their structure using NMR spectroscopy.Techniques and skills: Setting up reactions under anhydrous conditions, recrystallization, keeping a laboratory book, laboratory calculations, writing up materials and methods for publication. Planning, carrying out, troubleshooting and writing up experiments independently as part of an international research group.The aim of the project was to build on a library of antitrypanosomal heterocyclic compounds including benzothiophenes which have demonstrated significant in vitro activity and no toxicity to human cell lines.
