The goal of my work in the laboratory of Dr. Susan Taylor is to elucidate how the dynamic structure of cAMP-dependent protein kinase (PKA) and its interactions with associated proteins contribute to regulation and function. Given the dynamic nature of the holoenzyme complex during resting state and catalytic turnover, elucidation of conformational changes between enzyme states in the holoenzyme and validation that the holoenzyme in solution reflects the current crystallographic data are current challenges in the field. Utilizing small angle X-ray and neutron scattering (SAXS/SANS) along with segmental labeling, intramolecular contacts and conformational changes for various states of the enzyme would be mapped. In addition, SAXS and electron microscopy can be used to characterize much larger complexes than are typically possible with X-ray crystallography. The characterization of the interactions between the PKA and A kinase anchoring proteins (AKAPs) within large macromolecular signaling complexes and the effects that modification (ie. phosphorylation or mutation) cause on these structures are highly relevant to understanding how PKA signaling is regulated and how disease states occur.
