    Brief Synopsis of Project: CLN6 disease belongs to a family of rare, fatal neurodegenerative disorders collectively called neuronal ceroid lipofuscinoses (NCLs) for which no effective treatment currently exists. While affected cell types display accumulation of lysosomal storage material and neuronal degeneration, the mechanisms underlying this pathology is poorly understood. Animal models of CLN6 disease and human autopsies have revealed a correlation between regions of neuronal loss and activation of astrocytes and microglia, suggesting that gliosis may play a role in neuronal degeneration. The current project seeks to identify the role of gliosis in the progression of CLN6 disease. Cortical neurons and glia were harvested from Cln6-deficient and wild type (Cln6nclf and WT, respectively) mice at postnatal day 1 (P1) and postnatal day 2 (P2) for in vitro analyses. Utilizing ICC, I quantified the relative intensity of NCL hallmark markers (LysoTracker, ATP synthase subunit C, and autofluorescent storage material) in primary neuronal cultures. I have reported significant differences in NCL markers between Cln6nclf (designated Cln6 henceforth) and WT neuronal cells after maintaining cultures for 14 days in vitro (DIV14); decreased staining intensity of LysoTracker red and increased accumulation of ATP synthase subunit and ASM in Cln6 neurons was observed in comparison with staining intensities of WT neurons. To define the effect of Cln6- deficiency on the activation status of glia, I have isolated populations of resting astrocytes and microglia and immunostained them with activation markers glial fibrillary acidic protein (GFAP) and CD68, respectively. Resting Cln6 astrocytes and microglia displayed significantly higher intensities of glial fibrillary acidic protein (GFAP) and CD68 immunostaining, respectively, compared to resting WT glia, suggesting that Cln6-deficiency in glia predicts pre-mature activation.
