Dept. of Physiology and Biophysics, Advisor: Prof. Robert Chow.• Determination of the role of complexin II in regulated secretion from chromaffin cells using electrophysiology and a knockout mouse model. Significance: Understanding the protein machinery controlling exocytosis is of fundamental interest and could lead to more effective treatment of a wide variety of diseases, from Lou Gehrig’s disease to diabetes. Skills acquired: Electrophysiology and objective based Total Internal Reflection Fluorescence (TIRF) microscopy.

Dept. of Physiology and Biophysics, Advisor: Prof. William Betz.• Characterization of a concentration-dependent color shift of FM 1-43 stained granules in lactotroph cells. Significance: The color shift of FM 1-43 from green to red with increasing concentration could be used in a manner analogous to Fluorescence Resonance Energy Transfer (FRET) to monitor nanometer-scale distance changes between labeled moieties in live cells. Skills acquired: Mammalian cell culture techniques, spectral imaging, confocal imaging, MATLAB based image analysis.

Ph.D., Chemistry, Advisor: Prof. Steven Boxer.Dissertation title: “Investigations and Applications of Model Membrane Systems.”• Investigation of systems that mimic the cell membrane, including glass-supported lipid bilayers, reconstituted vesicles in solution, vesicles tethered to supported bilayers and DNA adsorbed to supported bilayers. Significance: Model membrane systems are useful for studying fundamental properties of cell membranes and membrane-protein interactions in a controlled environment. Furthermore, these systems are used in various bio-sensors for detection of membrane associated biomarkers. Techniques acquired: Formation of unilamellar vesicles, cleaning and smoothing of silica surfaces, poly-dimethyl siloxane (PDMS) based micro-contact printing, lipid biophysics, fluorescence imaging, atomic force microscopy (AFM), UV-VIS spectroscopy, optics.

Summer internship, Superconductivity Technology Center, Supervisor: Dr. Terry Holesinger.• Thermal processing and characterization of high temperature superconductors. Significance: The development of novel materials that super-conduct at liquid nitrogen temperatures (-70 K) will allow unprecedented improvement to the way our society transmits electricity. Skills acquired: Glove-box use for air sensitive materials, mixing and baking of materials to form ceramics in wires, scanning electron microscopy (SEM).
