Low bone density affects more than 40 million people and is a risk factor for osteoporotic bone breaks. In fact, 1 out of 2 women over the age of 50 and 1 out of 5 men will have an osteoporotic fracture in their lifetime. Bone breaks are painful, can take a long time to heal and sometimes result in permanent use of a cane or walker. Despite many available medications, the number of patients with osteoporosis and its associated fractures is increasing. To identify additional ways to enhance bone health, our lab is studying mechanisms of bone loss associated with diabetes, glucocorticoid use, estrogen-deficiency, antibiotic use, and inflammatory bowel disease. All of these conditions have a common link: dysbiosis (an imbalance of the gut microbiota). Our lab has shown that intestinal inflammation causes intestinal barrier leaks and bone loss. We have demonstrated that probiotics (bacteria beneficial to health) can increase bone density in healthy male mice and prevent bone loss in many conditions. This has led us to focus on the gut as a therapeutic target to treat osteoporosis. Using a variety of mouse and bacteria models we are working to understand how bacteria in the gut can regulate bone health. We are also studying the impact of changes to the microbiota on intestinal function (using barrier enhancing agents to prevent bone loss), bone cell transcription factor activity (Wnt10b signaling), intracellular signaling pathway activation, stem cell lineage selection, apoptosis and metabolism. Our studies, in collaboration with Drs. Parameswaran and Quinn, incorporate a team of scientists to better understand the microbiome, metabolome, mathematical modeling, bone strength and immune system contributions to the gut-bone signaling axis in animal models and clinically.I am an Associate Editor for the Journal of Cellular Biochemistry and serves on the Editorial Boards of Physiologic Reviews, Journal of Bone and Mineral Research and JBMR Plus. I also serves on national and international grant review panels including NIH, ADA, DOD. I am active in scientific societies and currently serve as the Chair of the American Physiology Society (APS) Science Policy Committee and serve on the FASEB Science Policy Committee, APS Council, Federal Demonstration Partnership Committees, and American Society of Bone and Mineral Research Mentoring team.
