At Brandeis University in the biochemistry laboratory of Gregory A. Petsko and Dagmar Ringe, my research involved elucidating the position of hydrogen atoms in the protein gamma-chymotrypsin at various pH values using neutron diffraction. This neutron data will then be correlated with a large amount of ultra-high resolution X-ray data in order to assess a geometric means of placing hydrogen atoms into proteins using only X-ray diffraction data. The goals of this project are to not only develop a standard for placing hydrogen atoms in a small subset of characteristic proteins, but also to develop computational methods of improving crystal structure accuracy as well as improving rational drug design techniques.

At Dickinson College in the plant biochemistry laboratory of Thomas Arnold, my project involved growing grape vines and treating the leaves with various agents such as jasmonic acid in an effort to increase the level of resveratrol in the vines. After the leaves had been treated with the jasmonic acid for several weeks, leaves were picked off the vines, frozen in liquid nitrogen, and then crushed and submitted to an HPLC for analysis. Using a resveratrol standard, we compared our standard to that in the leaves. At the Drexel University College of Medicine, in the biochemistry laboratory of Joseph Nickels, my research involved considering the effects on the Erg25 and Erg27 genes of Saccharomyces cerevisiae upon introduction of the drug Lovastatin, as well as to mutate various sterol response elements in Erg25 and consider the consequences. Used the beta-galactosidase assay to measure promoter activity. Discovered that blocking sterol synthesis results in the accumulation of sterol intermediates, as evidenced through the activation of various transcription factors.
