During my research visit abroad I have visited the Molecular Vaccinology laboratory headed by Prof. Denise Doolan.The goal of my visit was to identify a tentative set of defined pre-erythrocytic Plasmodium falciparum T-cell antigen targets that are recognized by volunteers who have been immunized with the ChemoProphylaxis and Sporozoites (CPS) regimen, in a preliminary proof-of-concept study. To this end, I have learned and utilized the interferon-gamma enzyme-linked immunospot (ELISpot) assay after re-stimulation of PBMC samples with peptide pools of P. falciparum antigens.Tasks: Isolation and cryopreservation of human materials, set up and optimizations of (cultured) enzyme-linked immunospots (ELISpots), in vitro re-stimulation of PBMC samples with peptide pools of P. falciparum antigens.
