Project: "Development of an Analytical Workflow for Ultraplexed Tissue via Reconstruct" Principal Investigator: Linnaea Ostroff Graduate Advisor: Gianna Raimondi This project aimed to examine a set of targets and correlate their expression to parvalbumin. This was undergone by analyzing ultraplexed tissue from rat basolateral amygdala (BLA). Evidence is converging to indicate that amygdalar inhibitory interneuronal circuitry plays an essential role in fear conditioning (Krabbe et al, 2017), and in many psychiatric disorders (Lüthi & Lüschler, 2014). This has implicated several calcium-binding proteins, but especially Parvalbumin (PV) as it the most prevalently expressed within the GABAergic interneuron population of the BLA (McDonald & Mascagni, 2001). Coexpression of PV and other proteins, RNAs, and extracellular structures are additionally relevant for the function of the BLA in fear learning. Perineuronal nets, an extracellular matrix that predominantly surround PV-expressing neurons, have been shown to be vital for the protection of fear memories in rodents (Gogalla et al, 2009). As anxiety-related behavior is recognized to modulate over the course of the estrous cycle of rodents, BLA inhibitory circuitry may be implicated as ovarian hormones have been shown to moderate PV expression as a function of activity (Blume et al, 2017). Due to the nature of Ultraplex, the resulting dataset is thousands of sections large. Therefore, this project reviewed the methodology proposed for the analysis of PV expressing neurons and their coexpressing markers.
