- Performed experiments to determine best treatment concentrations, cell seeding density for treatment, and treatment times to enhance antigen surface presentation for Mass Spec analysis- Started an R&D project to implement 3D cell culture methods. Experiments focused on optimization of methods, ensuring uniform culture methods, and how to scale up spheroids to large cell counts for Mass Spec analysis. Research Associate I
- Worked extensively with both suspension and adherent cancer cell lines, growing them up to high cell counts for Mass Spec.- Transfected plasmids into 293T cells to create a lentivirus stock for transduction into cell lines. These viruses ranged from HLA's to vaccine cassettes.- Transduced these viruses into cell lines of interest for cryopreservation and further testing. After selection, I would ensure proper expression within the cell line by qPCR.- Performed routine mycoplasma testing to ensure our cell lines remained contamination free.- Treated large quantities of cells to increase surface antigen presentation for Mass Spec analysis. Post treatment, large cell pellets were collected for Mass spec. Smaller pellets were collected for qPCR and NGS.- Completed these methods, transfection to cell pellet collection, for 10+ cells lines in one quarter- Performed data analysis on qPCR results- Presented cell culture workflow and results in group meetings-Assisted in the creation and optimization of Benchling ELN notebook and inventory
