Pdm Intern

Groton, Connecticut, United States

As an intern for the Pharmacokinetics, Dynamics, and Metabolism Department, I had the opportunity to delve into innovative research regarding models for preclinical efficacy. Working alongside experienced and supportive team members, I gained invaluable insights into this field, laboratory procedures, experimental design, and the importance of collaboration in scientific research. My primary project explored the OATP2B1 transporter and evaluating the clinical risk for significant drug-drug and food-drug interactions within the gut. I was tasked to characterize and optimize the substrate uptake conditions in a transporter assay at both intestinal and blood pH. I then used said conditions to evaluate 26 known transporter inhibitors and estimate their potency (IC50) using an inhibition assay. For those applicable, the data was then used in conjunction with clinical study data to create a static food-drug and drug-drug interaction assessment model. Finally, to conclude my project, I summarized and presented my findings via PowerPoint and poster presentations to the Pfizer scientific community. Throughout my internship, I am thankful to have gained hands-on experience in a variety of laboratory procedures, such as mammalian cell culture, kinetic analysis, literature search, ELN documentation, and LC/MS sample preparation, which greatly increased my technical abilities and skill-set. I was also fortunate to have been exposed to state-of-the-art equipment and software, including the VIAFLO 96-channel pipet. However, I would not have been able to have such success without the mentorship of the PDM team. Engaging with conversations, demonstrations, and departmental meetings allowed me to expand my knowledge, receive helpful suggestions, and develop myself as a upcoming scientist in pharmaceutical research.

Biotechnology Intern - In Vivo

New Haven, Connecticut, United States

I was very fortunate to have been invited back as an intern for Cybrexa Therapeutics, where I had the chance to continue to support preclinical Oncology projects for another summer. During the duration of this internship I experienced my first involvement in in vivo experimentation which allowed me to explore another aspect of pharmaceutical research and build my skill set to include working with small animals. My primary project was focused on establishing and executing methods to study immunogenic cell death (ICD) in tumors treated with chemotherapeutics delivered by a tumor-targeting peptide. I was again tasked with culturing tumor cells, which I then implanted into mice and monitored the growth of. In addition, I prepared dosing solutions and successfully dosed the mice. Furthermore, I harvested said tumors and digested them to prepare single-cell suspensions before staining for T-cell makers. I then analyzed the ex-vivo T-cell recruitment on an Accuri Flow Cytometer and, again, presented my findings at company meetings. Throughout my internship, I was informed of and practiced the safety and ethical protocols that are associated with in-vivo experimentation and small animal husbandry. In addition, I was able to learn and understand many new procedures such as implantation, dosing, and flow cytometry. I would again like to thank Cybrexa Therapeutics for their support, knowledge, and mentorship during my time as an intern!

Biotechnology Intern - In Vitro

New Haven, Connecticut, United States

As an intern for Cybrexa Therapeutics, I had the opportunity to support in vitro preclinical Oncology projects. This experience offered me a tremendous foundational knowledge of basic laboratory procedures and experimental design, as well as allowed me to gain my first hands-on and independent experience in a functional biotechnology laboratory. I owe my ever-growing passion for pharmaceutical research and development to the Cybrexa Team and their support during the duration of my internship. My primary project was focused on proof-of-concept experiments regarding a novel tumor-targeting peptide drug and it's ability to deliver chemotherapeutics directly to tumors based on pH. I was introduced to mammalian cell culture and tasked to grow, passage, and plate tumor cells for use in these experiments using proper aseptic technique. I also successfully prepared my own media at a variety of desired pHs. I then analyzed the pH-dependent uptake of LICOR-labeled alphalexTM and prepared drug-dose response curves using GraphPad Prism. Throughout the duration of my internship I would frequent company meetings where I engaged in conversations as well as presented my own findings for feedback. I am very thankful for the mentorship that the Cybrexa team offered me as I begun learning my way around a laboratory and the process of preclinical research. This experience offered me to cultivate my technical skills such as pipetting, media preparation, cell culture, and data analysis within a professional, yet supportive, environment.
