Assigned to Janssen to work on a PAT pilot project for Janssen R&D, involving the development and installation of a PAT system to enable automated feedback control of large molecule cell culture fermentation and small molecule chemical API synthesis. The PAT system will directly control (run, start, stop) PAT instruments (including Raman, FBRM and Mass Spec) and will also be able to model PAT data (spectral calibration models, MATLAB control models etc.) to generate predictions (process CQA's, chemical reaction endpoints, process step endpoints etc.) which can be written back to the PCS/DCS for process control and to the plant historian (OSI PI) for advanced data visualisation.

Title: Chemometric Modeling of Multiple Critical Process Parameters in Mammalian Cell Culture Bio-reactors using an Inline Raman Spectroscopy Probe.Working with Janssen Pharmaceutical Development and Manufacturing Sciences (large molecule), the aim of the project was to assess the use of in-line Raman data to model a mammalian cell culture process to support process understanding (FDA PAT initiative), and aid in process optimisation (feedback control).
