1. Over the course of three months I applied immunohistochemistry to marmoset, adult mouse, and mouse embryo brains in order to identify the best antibody combination for colocalizing axon pathways. This involved vibratoming the tissues into (typically) 200 um slices, potentially clearing the tissue, indirect IHC, confocal microscopy, and image analysis using ImageJ.2. Identifying which combinations of clearing technique (CLARITY, SHIELD, etc.) and IHC produced the strongest signals. With two others, using ImageJ on hundreds of images to determine what was signal (axons and soma) and what was noise (artifacts, blood vessels) by exporting the data to Microsoft Excel.3. Testing Janelia dyes for resistance to bleaching as an alternative to similar dyes.
