- Dissertation title: Ancestral sequence reconstruction as a lead optimization approach for gene therapy drug development.- Worked under the direction of Dr. Chris Doering utilizing ancestral sequence reconstruction to identify high activity coagulation FVIII and FIX variants to improve the potency of viral vector-based gene therapies for Hemophilia A and B.-First author publication work identified novel amino acid substitutions from computationally inferredancient FIX sequences that improve protein specific activity ~10-fold in vitro and in vivo. Theoptimized variant, termed ET9, has been licensed by Expression Therapeutics, with imminent plans for an IND-filing.
