Overexpressing the L-lactate-utilization lld operon in an attenuated tumour-targeting strain of SalmonellaThe aim of the project was to generate a strain of the attenuated Salmonella upregulated for utilisation of L-lactate. The bacterial lld operon, divorced from its native regulated promoters, was amplified and cloned after a strong constitutive tac promoter in a plasmid. L-lactate utilisation of the engineered Salmonella was then measured. Co-cultures of the bacteria with a breast cancer cell line, MCF7, was carried out and L-lactate produced was then quantified. These data informed on the likely ability of the bacteria to disrupt the balance of L-lactate in the TME and thereby suppress tumour growth.

Investigated the immune response in pregnancy, especially of placenta, to several antigens using ELISA and Western Blotting. Supervised by Professor Catherine Thornton.
